Transcription factor HERP1 regulates the target gene and monocarboxylate transporter 7 in human malignant melanoma.
The focus of our research is on the mechanisms involved in tumour formation and their chemo-resistance. The monocarboxylate transporter 7 (MCT7) is strongly overexpressed in metastatic human malignant melanoma (MM) and several proteins are responsible for its expression. We investigated whether HERP1, a known transcriptional repressor, is involved in MCT7 expression in melanoma cells. The transcription factor HERP1 was upregulated in melanoma cell lines and tissues and knockdown by RNA interference reduced MCT7 expression. We found that MCT7 expression was repressed by protein kinase A (PKA) in melanoma cells. HERP1 was found to be a protein substrate of PKA. Furthermore, HERP1 repressed MCT7 promoter activity, suggesting the involvement of the PKA pathway. PKA regulation by HERP1 on MCT7 expression was also supported by the fact that MCT7 overexpression led to decreased PKA activity. Moreover, HERP1 expression affected PKA-dependent cell growth, adhesion and motility in melanoma cells. Taken together, our data demonstrate that HERP1 is an important transcriptional regulator of MCT7 expression and that its expression is also associated with melanoma cell growth, adhesion and motility.In recent years, smart devices, such as smart phones, have become ubiquitous.
